dc.description.abstractИзучение особенностей токсического действия диизононилфталата на лабораторных животных показало инициирование тератогенных эффектов у крыс при внутрижелудочном введении самкам на протяжении периода беременности. Уровень воздействия 10000 мг/кг фталата характеризовался увеличением общей постимплантационной и эмбриональной смертности, наличием множественных (сочетанных) пороков развития эмбрионов. Постнатальное развитие потомства характеризовалось нарушением функции эндокринной системы: щитовидной и половых желез, приводящим к развитию первичной тестикулярной недостаточности и тиреотоксикозу. В эксперименте установлена максимально недействующая доза 10 мг/кг. Элементами механизма эмбриотоксического действия диизононилфталата являются нарушения процессов клеточной дифференцировки, ДНК-повреждающее действие и цитотоксические свойства фталата, которые приводили к образованию полиплоидных клеток в селезенке при однократном внутрибрюшинном воздействии фталата в опытах на белых мышах, и к накоплению лейкоцитов периферической крови с нарушениями генетического аппарата и дифференцировки (накопление сегментоядерных клеток, микроядер, клеток с признаками некроза) при субхроническом внутрижелудочном введении самцам белых крыс.
dc.description.abstractThe study of the peculiarities of diisononyl phthalate toxic effects on laboratory animals showed the initiation of teratogenic effects in rats on intragastric administration to females during the gestation period. The exposure level of 10,000 mg/kg of phthalate was characterized by an increase in total postimplantation and embryonic mortality, the presence of multiple (combined) malformations of embryos. Postnatal development of the offspring was characterized by the disturbance of the endocrine system function: the thyroid and sex glands, leading to the development of primary testicular insufficiency and thyrotoxicosis. In the experiment, the most inactive dose was 10 mg/kg. Elements of the embryotoxic action mechanism of diisononyl phthalate are the disturbances in the processes of cell differentiation, DNA damaging effect and cytotoxic properties of phthalate, which led to the formation of polyploid cells in the spleen on single intraperitoneal impact of phthalate in experiments on white mice, and accumulation of peripheral blood leukocytes with disorders of the genetic apparatus and differentiation (accumulation of segmentated cells, micronuclei, cells with the signs of necrosis) on subchronic intragastric introduction to male white rats.
