Abstract: Цель работы – изучить влияние природы и концентрации (объемной доли) органических экстрагентов на извлечение флавоноидов из календулы цветков. Объектом исследования служили календулы цветки промышленной серии. Материал и методы: высокоэффективная жидкостная хроматография. Перечень используемых в работе органических растворителей приведен ниже. Изучали следующие объемные доли этих растворителей: 20, 40, 60, 80 и 100%. Результаты. В статье представлены результаты изучения экстракции флавоноидов из календулы цветков метанолом, этанолом, пропанолом-1, ацетоном, диметилсульфоксидом и их водными растворами, а также водой, пропанолом-2, бутанолом-1, бутанолом-2, этилацетатом. В полученных извлечениях при помощи метода высокоэффективной жидкостной хроматографии обнаружены от одного до восьми флавоноидов, преобладали производные изорамнетина. Экстрагирующая способность абсолютных экстрагентов в отношении флавоноидов календулы цветков уменьшалась в ряду: вода> метанол> диметилсульфоксид> этанол> бутанол-1> этилацетат> бутанол-2> ацетон> пропанол-1> пропанол-2. Экстрагирующая способность пропанола-1 и ацетона уступала их водным растворам. Экстрагирующая способность метанола, этанола и диметилсульфоксида сопоставима с их водными растворами. Диэлектрическая проницаемость, динамическая вязкость, температура кипения, объемные доли экстрагентов в растворах статистически значимо (p<0,05) влияли на площади хроматографических пиков флавоноидов. Наиболее критическим фактором являлась объемная доля этанола в растворе. Для дальнейшего количественного определения флавоноидов в календуле цветках, учитывая значительную экстрагирующую способность и хорошую воспроизводимость результатов количественного определения, рекомендовано использовать 60% ацетон. Заключение. Изучена изменчивость качественного и количественного состава извлечений из календулы цветков при экстракции различными экстрагентами.
Objectives. To study the influence of nature and concentration (volume proportions) of organic extractants on the extraction of flavonoids from calendula flowers. Material and methods. The object of the study was calendula flowers of the industrial series. The method used in the investigation was high-performance liquid chromatography. The list of organic solvents used in the work is given below. The following volume proportions of these solvents were studied: 20, 40, 60, 80 and 100%. Results. The results of the study of flavonoids extraction from calendula flowers with the use of methanol, ethanol, propanol-1, acetone, dimethylsulfoxide and their aqueous solutions, as well as water, propanol-2, butanol-1, butanol-2, ethyl acetate are presented in this article. From one to eight flavonoids were detected in the prepared extracts by the method of high-performance liquid chromatography, isoramnetin derivatives predominated. The extracting ability of absolute extractants with regard to flavonoids of calendula flowers decreased in the series: water> methanol> dimethylsulfoxide> ethanol> butanol-1> ethyl acetate> butanol-2> acetone> propanol-1> propanol-2. The extracting ability of propanol-1 and acetone was lower compared with that of their aqueous solutions. The extracting ability of methanol, ethanol and dimethylsulfoxide was comparable with that of their aqueous solutions. Dielectric permeability, dynamic viscosity, boiling point, volume proportions of extractants in solutions statistically significantly (p<0,05) influenced the area of chromatographic peaks of flavonoids. The most critical factor was the volume proportion of ethanol in the solution. It is recommended to use 60% acetone for further quantitative determination of flavonoids in calendula flowers taking into account the significant extractive capacity and good reproducibility of the quantitative determination results. Conclusions. The variability of qualitative and quantitative composition of extracts from calendula flowers was studied during extraction by means of various extractants.
