Abstract: Целью данного исследования было изучение полисахаридов пармелии слоевищ. Материал и методы. Объектом исследования служили измельченные пармелии слоевища. Экстракцию суммы водорастворимых полисахаридов из сырья проводили горячей водой. Для выявления полисахаридов использовали реакцию осаждения 96% этанолом. Количественное определение суммы водорастворимых полисахаридов проводили гравиметрическим методом. Из обезжиренного шрота выделяли отдельные фракции полисахаридов. Фракцию водорастворимых полисахаридов (ВРПС) получали экстракцией горячей водой. Пектиновые вещества (ПВ) извлекали смесью 0,5% растворов кислоты щавелевой и аммония оксалата (1:1), а гемицеллюлозу А (ГЦ А) и гемицеллюлозу Б (ГЦ Б) – 7% раствором гидроксида натрия. Результаты. При добавлении к 3-кратному объему 96% этанола водной вытяжки пармелии слоевищ образовывался коричневый аморфный осадок, что свидетельствовало о присутствии полисахаридов в исследуемой вытяжке. В результате количественного определения суммы водорастворимых полисахаридов было установлено, что их содержание в пармелии слоевищах составило 10,95 ±0,35%. При фракционировании полисахаридов выход фракции ВРПС составил 8,90±0,30%, ПВ – 12,32±0,42%, ГЦ А – 14,67±0,47%, ГЦ Б – 8,47±0,28%. Заключение. В результате проведенных исследований было установлено содержание суммы водорастворимых полисахаридов пармелии слоевищ. Впервые выделены отдельные фракции полисахаридов и определено их количественное содержание. Полученные результаты свидетельствуют о перспективности дальнейшего изучения полисахаридных комплексов пармелии слоевищ с целью создания новых эффективных фитосредств.
Objectives. To study polysaccharides of Parmelia thalluses. Material and methods. Crushed Parmelia thalluses were the object of the study. The extraction of the sum of water-soluble polysaccharides from the plant raw material was carried out by hot water. Precipitation reaction by 96% ethanol was used to detect polysaccharides. Quantitative determination of the sum of water-soluble polysaccharides in Parmelia thalli was made by the gravimetric method. Separate fractions of polysaccharides were isolated from the defatted extraction cake. The fraction of water-soluble polysaccharides (WSPS) was obtained by hot water extraction. Pectin was extracted using the mixture of 0,5% solutions of oxalic acid and ammonium oxalate (1:1), and hemicellulose A (HC A) and hemicellulose B (HC B) – by means of 7% solution of sodium hydroxide. Results. On adding to the aqueous extract of Parmelia thalluses the threefold volume of 96% ethanol the brown amorphous precipitate formed, which confirmed the presence of polysaccharides in the studied extract. As a result of the quantitative determination of the sum of water-soluble polysaccharides it was found, that their content in Parmelia thalluses made up 10,95±0,35%. On fractionation the yield of different fractions of polysaccharides was the following: WSPS – 8,90±0,30%, pectin – 12,32±0,42%, HC A – 14,67±0,47%, HC B – 8,47±0,28%. Conclusions. The content of the sum of water-soluble polysaccharides in Parmelia thalluses was determined as a result of the conducted studies. For the first time individual fractions of polysaccharides were isolated and their quantitative content was determined. The results obtained testify to the prospects of further study of polysaccharide complexes of Parmelia thalli in order to create new effective phytoremedies.
